Electroporation optimal conditions for Endura cells

Can any one please please help me, I am struggling to find a protocol to use with the Endura cells for electroporation with the GeCKO libraries. I am following the guidelines set out by the Lucigen company but pulse length, numbers and exponential decay are not taken into consideration.

I would really appreciate any help.

Optimal conditions are specified in the Endura manual: http://lucigen.com/docs/manuals/MA133-Endura-Competent-Cells.pdf

I amplified mouse GeCKO library plasmid recently according to manufacturer's instruction which Ophir has attached.

I used 2mm cuvette (Biorad) at 25uF, 200 ohms and 2.5 KV. I got 10-20 times more colonies than recommended colony number in the GeCKO library instruction sheet.

Hope it helps.

It is interesting to see your condition get more colonies. Have you compared the condition you used with that in Gecko library instruction sheet side by side? Or, did you just compare the colony number with 3 million?
