Clonogenic Assay (Colony Formation Assay)

One way to determine the effect of treatments (e.g. drugs) on cell proliferation and survival is to work with the Colony Formation Assay.

I seeded 1000 cells (HeLa) per well and incubated them with fresh medium after treatment (e.g. Irradiation of 2 Gy) for around 10 days. After that, I stained cells with crystal violet to count them (compared to untreated/unirradiated cells). You calculate the survival fraction and plot it easily to see if your data is significant. What I like about this assay: It is extremely easy to perform and while waiting for 10 days you have time for other things. Even though, the technique is not time consuming at all, I noticed that it provides valuable information.
